Han and colleagues (2003) have developed various retinol derivatives to improve the photostability of retinol while retaining its anti-aging activity. A: The discoloration (turns brown) of the Lightening Gel is a normal reaction of this formula due to exposure to air and/or high temperature. Alirezai and colleagues (1994) conducted a vehicle-controlled study which involved the use of 0.1% isotretinoin cream twice daily for 24 weeks. The study resulted in a statistically significant reduction in actinic keratoses and precancerous facial lesions in the isotretinoin group, with 66% of 44 patients achieving a reduction in more than one-third of lesions. However, no significant drug effect was seen for actinic keratoses on the scalp or upper extremities. Mild-to-moderate irritant reactions were observed in the isotretinoin treated group, but symptoms subsided with reduced frequency of the treatment.

Nanoparticles are extensively being investigated for drug delivery in the pharmaceutical research from the last 3 decades on. Nanoparticles offer immense benefits such as solubilization of hydrophobic actives, improvement in bioavailability, improved (or altered) pharmacokinetics of active pharmaceutical ingredient (API), protection of API from physical, chemical or biological degradation, improved cellular uptake, tissue targeting and controlled release of API. However, most of the initial investigations on nanoparticles dealt with the parenteral or oral delivery of APIs whereas efforts to explore the potential of nanoparticles in topical delivery were initiated in the last decade. Interestingly, nanoparticles were found to improve the distribution characteristics and stability of topically applied APIs such as sunscreens (Wissing and Muller 2002).

Hence, in order to minimize or avoid these disadvantages, Kligman and colleagues (1998) evaluated the potential of high strength tretinoin (0.25% solution in a fast penetrating vehicle) for the treatment of photoaging in 50 females. The treatment regimen consisted of application of highly concentrated tretinoin solution on alternate nights for 2 weeks and then every night thereafter until the end of the treatment. Interestingly, just 4 to 6 week treatment with high strength tretinoin resulted in improvement in fine wrinkling, mottled hyperpigmentation, elasticity, hydration, angiogenesis, and new collagen deposition above the zone of solar elastosis and the extent was similar to the results observed after 6 to 12 months of standard tretinoin therapy (0.05%). Moreover, the high strength tretinoin treatment was well tolerated in all patients. Subsequently, Cuce and colleagues (2001) evaluated efficacy of the 1% tretinoin solution applied twice a week in 15 women with photodamaged skin. Histological studies carried out after 15 days showed compaction of stratum corneum and increased epidermal thickness. Additionally, surface imaging studies showed improvement in skin texture and appearance.

Furthermore, a wide band of eosinophilic material just beneath the epidermis, devoid of oxytalan fibers and forming the ‘grenz zone’, appeared more frequently and was larger in the retinol- and vitamin C treated group.

Development of receptor selective synthetic retinoids (like seletinoid G) or novel retinoid derivatives (like N-formyl aspartamate derivative of retinol) or retinoid co-drugs like retinyl ascorbate (Abdulmajed and Heard 2004) which may be superior in terms of tolerability, stability and efficacy.

Quick Summary:  Kate Somerville Retasphere Retinol Night Cream is a very good retinol cream.  We saw pretty good results both in the short term and longer term when using this product (especially on fine lines and increasing skin smoothness).  It’s also very moisturizing– which is something you can’t say about all retinol creams!  As far as downsides, this product is definitely on the thicker side, which some of you will not like. I love the DDF Amplifying Elixir. It cost $70 and you can buy it from most upmarket department stores. This product contains Niacinamide which is proven to even out skin tone and fade spots. The only problem I have with DDF is they keep discontinuing and replacing products so if you are using a cleanser, serum or cream you love it might disappear and be replaced by something that just doesn’t work for you.

When it comes to Byrdie-editor favorites, one product seemed to keep popping up: The Kate Somerville Retinol Firming Eye Cream. It goes on pretty lightweight and comes with a cooling gold applicator that just feels so nice. Most of us who tried it definitely noticed a difference after a couple of uses.

This serum has a thicker consistency than most other eye serums I have tried. It applies smoothly and dries completely within about 4 minutes. Moisturize your skin regularly when using retinol. Apply extra moisturizer after you have used your retinol cream. Although it is good to use for sun exposure, it can also lead to sun sensitivity.

Bhawan and colleagues (1996) studied the effect of long-term use (4 years) of tretinoin emollient cream in 27 patients treated with either 0.05% or 0.01% of tretinoin for the first 18 months, followed by 15-month treatment with 0.01% tretinoin and finally 19-month daily treatment with either 0.025% or 0.05% tretinoin. Histological studies indicated that the stratum corneum became compact in the first 3 to 6 months whereas it returned to normal (basket weave pattern) in 12–24 months and remained normal until the end of the therapy. Likewise, granular layer thickness and epidermal thickness were increased in the first 3–6 months, returned to normal in 12–24 months and remained normal until cessation of the therapy. In contrast, epidermal mucin continued to increase and melanin continued to decrease throughout tretinoin treatment. The changes in these 2 components clearly correlated with the observed clinical changes.

Though you can ask your doc for a prescription-strength retinoid—which is a much stronger version of what you can buy over the counter—the derms we spoke to said drugstore and beauty counter brands do the same thing, and that it just may take longer to see results. To date only one vehicle-controlled clinical study has been undertaken to evaluate the use of topical tretinoin for the treatment of chronologically aged skin. Ultrastructurally, an increase in anchoring fibrils was noted at the level of the dermo-epidermal junction. In the dermis, development of several new micro vasculatures (angiogenesis) and production of new elastic material and GAGs was observed. These morphological changes suggested that the magnitude of effect of tretinoin might be greater for chronologically aged than for photoaged skin. However, large scale, multicentric clinical trials need to be conducted for confirming the utility of tretinoin for chronological aging.

Significant improvement in histological and clinical markers was observed in both the 0.05% and the 0.01% tretinoin group as compared with vehicle. In another study, Oslen and colleagues (1997b) evaluated the 6 month effect of once weekly or thrice weekly 0.05% tretinoin emollient or no treatment in 126 individuals who had completed 48 months of 0.05% once daily tretinoin therapy. Thrice weekly tretinoin treatment appeared to be more effective in improving the fine wrinkles than once weekly therapy whereas discontinuation of the therapy resulted in the reversal of beneficial effects to some extent.

For first-timers, over-the-counter (OTC) formulations, like the dermatologist-recommended products listed on the next page, are the mildest and the best way to start. “The most common retinoids on these labels are retinol and retinyl palmitate,” says Chris R. Retinol is a specific form of vitamin A that is naturally produced in your body. When it’s applied topically, enzymes in your skin convert retinol into retinaldehyde and then into retinoic acid. I think im allergic to retinol. Would that be possible ? I had good skin and decided to start retinol to prevent aging (im 25). I used peter thomas roth retinol fusion pm for 3 1/2 months and developed the worst cystic acne ive even seen. I now have pimples all over my cheeks and chin (eeeeeverywhere). Its all red and swollen, you cant see skin underneath. I stopped using the profuct 2 weeks ago and my skin has calmed quite a lot. But i started using roseuip seed oil and cystic acne seems to reach the surface again, tho not as intensely as with the retinol i was using.

i have to say I wanted to love this eye cream, so I ve been using it for more than 6 months. I do not have wrinkles but I have dark circles under my eyes. It stings a little bit. Retinoic acid via the retinoic acid receptor influences the process of cell differentiation, hence, the growth and development of embryos. During development there is a concentration gradient of retinoic acid along the anterior-posterior (head-tail) axis. Cells in the embryo respond to retinoic acid differently depending on the amount present. For example, in vertebrates the hindbrain transiently forms eight rhombomers and each rhombomere has a specific pattern of genes being expressed. If retinoic acid is not present the last four rhombomeres do not develop. Instead rhombomeres 1–4 grow to cover the same amount of space as all eight would normally occupy. Retinoic acid has its effects by turning on a differential pattern of Homeobox (Hox) genes which encode different homeodomain transcription factors which in turn can turn on cell type specific genes. Deletion of the Homeobox (Hox-1) gene from rhombomere 4 makes the neurons growing in that region behave like neurons from rhombomere 2. “Retinol Cream Loreal +Retinol Vitamin-A Retinol Tretinoin Cream”

I use a number of Roc products & as usual, this one works as well as the other Roc products. This one claims: • Moisturizes and visibly reduces signs of aging around the eyes • Reduces appearance of crow’s feet, dark circles, and puffiness • Formula contains retinol and exclusive mineral blend • Clinically shown to reduce wrinkles around the eyes in 12 weeks While I haven’t used it for 12 weeks yet, I have not seen the full results. What I have noticed is that the dark circles have lightened up. It hasn’t reduced the puffiness under my eyes yet, but I have noticed a slight reduction in the wrinkles around my eyes. This is not greasy or sticky & I have had bad effects of the Retinol in this cream. I’m hopeful that when I have used this for a full 12 weeks, I will have better results. This retinol cream is a little richer and more velvety than the others – perfect if you like lots of moisture. For those whose skin isn’t accustomed to retinol, it comes with a ‘Buffer Cream’ to make it more tolerable. Simply mix one part retinol and three parts buffer and apply it directly onto cleansed skin at night. If you don’t experience any irritation, you can slowly wean yourself off of the accompanying cream. It’s brilliant at evening out skin tone and texture. 

I hope Amazon starts carrying this product again soon… I have been acne free (minus a few hormonal zits that clear up quickly) for 6 months straight. This has not happened in at least 10 years! This product works! I’m on my third bottle. Well worth the price.

This is probably one of the worst eye creams I have ever used. It is supposed to provide moisture and help lessen fine lines and wrinkles. All this did is dry up my eyes and provide more wrinkles. It burns and sting my eyes. Probably this is not suitable for those with sensitive and thinner skin
